Are Rab guanosine triphosphatases involved in the formation as well as the targeting of transport vesicles containing protein cargo? In this Perspective, Segev explains new findings showing that this is indeed the case. Not only does Rab9 recruit its effector TIP47 to budding vesicle membranes, it also enhances interactions between TIP47 and the mannose-6-phosphate cargo receptor. A complex among Rab9, TIP47, and mannose-6-phosphate receptors is probably important for vesicle budding.
