If the precise level and/or location of drug in the body could be controlled, side effects and required doses could be reduced and new therapies would become possible. In his Perspective, Langer describes advances toward this goal, from gene therapy to oral and pulmonary delivery of drugs. Many challenges remain, but recent advances promise highly targeted drug delivery, with major benefits to patients.
