Regulating the expression of target genes in response to different conditions is one of the most daunting tasks facing the cell. In a Perspective, Struhl describes the intricate steps involved in assembly of a precise transcriptional on-off switch called the enhanceosome, which regulates the transcription of the interferon-b gene in response to viral infection ( Munshi et al.).
