An essential element of cell survival is the ability to respond to DNA damage. In his Perspective, Carr discusses new findings that reveal the molecular underpinnings of one of the checkpoint pathways that recognizes DNA damage and arrest of DNA replication. This pathway is activated by checkpoint proteins binding to single-stranded DNA that is bound to the RPA protein (Zou and Elledge).
