Given that 80% of patients with type 2 diabetes are overweight, there must be a molecular link between obesity and diabetes. In their Perspective, Muoio and Newgard, discuss a new study ( Özcan et al.) that dissects the molecular connections between signaling pathways controlling fat metabolism and insulin action. It turns out that obesity puts extra stress on the endoplasmic reticulum, which initiates a series of responses that result in blocking of the insulin signaling pathway.
