Custom Antibody Panels

Custom Design Services provides researchers with single and multicolor custom-tailored conjugate configurations as well as purified antibodies. Two- to 10-color panels can be composed using fluorochromes to meet specific experimental requirements. Proprietary tandem dye technology provides optimal coupling efficiency and enhances signal-to-noise ratio, so that dyes can be used to detect low-density antigens without the loss of sensitivity. Antibodies may be lyophilized or supplied in a specified buffer solution. Krome Orange, a novel violet-excited organic dye, expands the range of available choices and dye sensitivity limits for violet lasers and facilitates the development of high-quality, multicolor panels. The panels are packaged in exclusive black vials to protect against photobleaching. The conjugation processes are controlled to provide optimal coupling efficiency, and lower compensation requirements simplify instrument setup and give more flexibility in optimizing multicolor combinations.

STR Profiling Service

The Short Tandem Repeat (STR) Profiling Service offers investigators an easy and sure solution for verifying the identity of their human cell lines. Researchers simply apply cells to an FTA card following the instructions provided with the FTA Sample Collection Kit, eliminating the need to ship samples on dry ice. ATCC will amplify the DNA using the Promega PowerPlex 16 HS System and analyze the results. Investigators can expect results via e-mail within five days from receipt of their sample. Two levels of service are offered: Basic and Verified. The Basic STR Profiling Service (135-X) includes a signed Cell Line Authentication Report containing an easy-to-understand STR allele table and electropherograms that support the allele calls at each locus. The Verified STR Profiling Service (135-XV) includes everything in the Basic service, plus known reference profiling against the ATCC STR Profile Database, and a comprehensive interpretation of the results.

UbiquitinProfiler Services

The UbiquitinProfiler Services determine the potency and selectivity of compounds targeting ubiquitin ligases, key enzymes in a pathway linked to many serious diseases. Through this service, compounds are assessed via a panel of validated assays for their ability to enhance or inhibit the ubiquitination pathway. With a central role in trafficking proteins for degradation, the ubiquitin pathway is an attractive target for drug discovery efforts in many areas including neurodegeneration, cancer, and inflammation. With hundreds of proteins involved, however, the complexity of the ubiquitin cascade has presented a formidable barrier to efficient drug discovery. With UbiquitinProfiler Services, the ways in which potential drug compounds impact the ubiquitin pathway can be more rapidly and effectively assessed. Previously, researchers had to develop and optimize all the necessary reagents and assays themselves.

RGS Protein Screening

A new high throughput screening (HTS) assay is available for screening the "Regulators of G-protein Signaling" (RGS) proteins that influence signaling of G Protein-Coupled Receptors (GPCRs). Offered as an assay development service, the RGScreen Assay Service opens up new therapeutic strategies for modulating GPCR pathways, including the potential to fine-tune the effects of existing GPCR ligands. RGS proteins attenuate GPCR signals by increasing the GTPase activity of associated Gα proteins. Though RGS proteins have been the focus of intense investigation as potential drug targets, the lack of suitable HTS assay methods has prevented large-scale screening efforts. BellBrook overcame this technical hurdle by developing proprietary Gα variant proteins that enable direct detection of RGS catalytic activity using their Transcreener HTS Assay platform. The RGScreen Assay Service provides custom assay development to produce and validate desired RGS protein targets followed by ongoing reagent supply, including RGS proteins, variant Gα proteins, and Transcreener GDP Assay components, to support large-scale RGS protein screening programs.

Protein Variant Screening

The SURE Variant Screening Service accelerates and improves outcomes in display library screening campaigns by reducing the number of steps and increasing the total productivity of the system. This service utilizes the proprietary Selexis SUREtechnology Platform and the Selexis SURECHO-M cell line to generate variant panels ranging from several to >100 stably-expressing protein variant cell lines, with typical expression levels above 100 mg/L. These variant proteins, expressed with mammalian modifications, can be assessed for activity. Manufacturing cell lines of the top candidates can be readily isolated from the stable transfectants. The Selexis SURE Variant Screening applied to protein display screening campaigns: (i) determines the values of mammalian protein modifications early; (ii) weeds out candidates that cannot be easily expressed in mammalian cells; (iii) ensures a steady supply of preclinical material; (iv) promotes faster, more informed decision-making; (v) eliminates the need for transient transfection; and (vi) significantly reduces development time.
