  • Movie S1
    PANC-1 ICAs formed during 16 days in SFM were exposed to SCM and observed by time-lapse photography every 5 min for the next 18 hours. Differentiated PANC-1 cells in the ICAs migrated out of clusters to form flattened cell monolayers that appear similar to the original undifferentiated PANC-1 cells in SCM. Total length, 35 seconds.

    PANC-1 ICAs were exposed to SCM for 12 hours, fixed in 4% paraformaldehyde and immunostained for insulin (red), Pdx-1 (Blue) and Ck-7/19 (green) as described in the Methods. This movie shows immunostaining in z-stack at different optical slices scanned using a confocal laser scanning microscope. Pdx-1 was observed primarily within the ICAs. Cells migrating out of the ICAs mostly express Ck-7/19 (green) and about 10% co-express insulin, visualized as yellow. Each optical slice, 0.9 μm.
