Two-photon-GRIN-lens volumetric imaging of PVH neuron calcium activity during fear retrieval experiment. PVH neuronal ensemble activity and mouse behaviors during Baseline, Eat-Hed, Fear, Tone-Aft and Spout-Aft states in one trial of the fear retrieval experiment. Top 2 rows show the volumetric calcium imaging of PVH neuron activity. ΔF/F activity (black-to-yellow colormap) is superimposed (alpha = 0.5) with the template reference images (gray) from 8 imaging planes. ΔF/F was calculated for each pixel's time series as previously described (65) with minor modification (see Methods). The template reference images are the average intensity projection of the temporal calcium images. To clearly show all neurons in the template images, Gamma of the template images was adjusted to 0.45. Bottom row shows the video of the mouse behaviors (Left: front view; Right: side view). The ensemble activity patterns are visually distinct in different states. The red box signifies the auditory cue (conditioned with electrical foot-shock in a foot-shock arena on the day before the experiment—no shock during the imaging experiment), and the mouse immediately stops licking. Lick suppression continues after the offset of the tone (Tone-Aft state). Video was played back at 6× speed. Audio was played back at 1× speed but trimmed to video playback speed.
